1. Push the syringe piston in fully. Place only thin part of the capillary into the bottom of the sample while angling the sample container at about 45 degrees. (Picture 1)
2. Placing two fingers below the piston head pull the piston back slowly while keeping the tip of the capillary well below the sample level and below any surface bubbles. Continue to aspirate the sample until it appears in the Luer adaptor.
3. Holding the capillary vertically, visually confirm that the sample has completely filled the thin section, the cuvette section and appears in the Luer adaptor. Tap on the syringe to make sure there are no air bubbles in the sample.
4. Quickly (to avoid wicking) and thoroughly wipe the outer surface of the capillary (Picture 2) - both top and bottom - removing all semen from the exterior of the capillary in order to prevent the SQA-V optical chamber from becoming clogged.
4. Slowly and carefully push-in the separating valve until it is level with the plastic (Picture 3). The capillary is now ready to be inserted into one of the SQA-V compartments for testing or viewing.
6. Firmly push the capillary into one of the compartment slots as far as it will go. Ensure that the capillary is properly seated and stable in the compartment - this may require a little extra pressure.

SQA-V Technology
Automated Testing System
Step 1: The capillary is inserted into the measurement compartment.
Step 2: Sample concentration is evaluated in the “tall” 10 mm chamber of the capillary by measuring the amount of optical absorption of light as a beam traverses the seminal fluid.
Step 3: Motility is detected in the “thin” 0.3 mm section of the capillary by analyzing light modulations caused by the sperm motion.
Step 4: This information is then digitized and routed to the microprocessor that applies algorithms to extract the required clinical semen parameters.
Motility Channel
Light disturbances caused by moving sperm cells are detected and translated into analog signal peaks.
The greater the number of motile sperm cells in the field of view, the higher the peaks.
The slower the sperm velocity is, the broader the signal peaks will be.
The signals are averaged and translated into MSC (Motile Sperm Concentration).
Concentration Channel
Sperm concentration is measured in the cuvette section of the SQA-V testing capillary.
An infrared light of a specific wavelength that is optimally and specifically absorbed by sperm cells and minimally absorbed by other seminal fluid components is produced by the SQA-V.
In the final calculation of Sperm Concentration, the SQA-V algorithm reads the light absorbed by sperm cells and makes a correction for the light absorbed by the seminal plasma components in order to accurately report sperm concentration.
Visualization System
A standard microscopy slide placed in the SQA-V slide adaptor can be inserted into the video (upper) screen of the SQA-V to view “real time” semen samples.
SQA-V capillaries can also be inserted into this chamber for “scanning” the sample.

Concentration
Millions of sperm cells are analyzed: A very specific wavelength of light is absorbed by the sperm cells in the concentration chamber of the SQA-Vt testing capillary. An optical density detector measures the light absorbed by the cells. This is translated into cell concentration by a microprocessor based on a proprietary MES algorithm.
Motility
Tens of thousands of sperm cells are analyzed as they move through a light source in the SQA-Vt: The movement of motile sperm causes light disturbances. These light disturbances are converted into electronic signals with “peaks and valleys.” These electronic signal peaks are analyzed by a microprocessor and translated into motility based on a proprietary MES algorithm.
